Redox-mapping MRI in rodent brain with nitroxide contrast agents

INTRODUCTION Overproduction of reactive oxygen species (ROS) and decreased antioxidant defense system may contribute to numerous brain disorders such as stroke, epilepsy, Parkinson’s disease, and Alzheimer’s disease. Non-invasive evaluation and visualization of oxidative stress is important to elucidate the role of ROS in brain diseases. In order to visualize the effect of ROS in vivo, nitroxid...

An overview of responsive MRI contrast agents for molecular imaging.

This review focuses on MR contrast agents that are responsive to a change in physiological environment. The "response" mechanisms are dependent on 6 physicochemical phenomena, including the accessibility of water to the agent, rotational tumbling time, proton exchange rate, electron spin state, MR frequency, or local field inhomogenieties caused by the agent. These phenomena can be affected by ...

Responsive paramagnetic chemical exchange saturation transfer MRI contrast agents

Molecular imaging research has shown great potential to assess the molecular compositions of tissues and provide critical information for clinical diagnoses [1]. These noninvasive assessments of molecular compositions may provide earlier diagnoses of pathologies, may predict therapeutic effects before the therapy is initiated and may provide earlier assessments of therapeutic effects soon after...

Redox-sensitive contrast agents for MRI based on reversible binding of thiols to serum albumin.

DOTA-based complexes of gadolinium (Gd) bearing a thiol moiety on a propyl or hexyl arm were synthesized. It was hypothesized that these complexes would form reversible covalent linkages with human serum albumin (HSA), which contains a reactive thiol at cysteine-34. The binding constant of the hexyl complex to HSA was measured to be 64 mM(-1) and decreased to 17, 6.1, and 3.6 mM(-1) in the pres...

Highly relaxing gadolinium based MRI contrast agents responsive to Mg2+ sensing.

A Gd complex based on a polyphosphonated pyridyl ligand shows a very high stability in aqueous solution (log K(EuL) = 25.7), a high relaxivity (8.5 mM(-1) s(-1) at 25 °C and 20 MHz) and a marked and selective relaxivity enhancement (37%) in the presence of Mg(2+), opening interesting perspectives for the design of cation responsive contrast agents.
